The 4" is a whale of a lot better to wear on a belt. The 6" will (probably) give you a little more velocity, and it'll be easier to shoot accurately due to the longer sight radius.
But if it were my choice, I'd go 4" every time. Oh, the GP100 is one of the finest revolvers ever made. Hell for tough like all Rugers, easy to smooth up if you need that, and warrantied forever.
